The cyclic interconversion of cell signaling proteins between modified forms and unmodified forms is critical for many important biological processes in vivo. The cyclic oxidation and reduction of methionine(Met)containing proteins play an indispensable role in controlling homeostasis in biological systems and regulating cellular signal transduction. In this work,a new cancer biomarker LAPTM4B protein was employed as a model protein to study the cyclic oxidation and reduction process of an extracellular fragment EL2 in vitro. To mimic the biological oxidation condition, hydrogen peroxide was used as the reactive oxygen species to oxidize the peptide.
