The frequently observed hypoxic microenvironment inside solid tumors including hepatocellular carcinoma (HCC) orchestrates a wide spectrum of molecular pathways that frustrate therapy.The hypoxic response is governed by hypoxia-inducible factors (HIFs), each of which is a heterodimer composed of α-subunit and *3f-subunit.HIFs regulate a wide range of hypoxia-related molecules control the expression of hundreds of genes, which determines cell proliferation, apoptosis, tumor progression and metastasis.The discovery of HIF-1 α subunit was followed by the discovery of the HIF-2α and HIF-3α subunits.Although HIF-3α is estranged from the other two subunits, both HIF-1α and HIF-2α activate transcription of hypoxia-inducible genes in an HRE-dependent manner.
