Objective: To investigate whether the lung cancer cell can down regulate the expression of IFN-γ gene in CD4+T cells directly through hypermethylation.Methods: Level of plasma IFN-γ was determined by ELISA in lung cancer patients group (n=30) and healthy controls group (n=30).CD4+T cells were isolated by CD4-positive isolation kit from peripheral blood of lung cancer patients (n=1 1) and healthy controls (n=10), genomic DNA was extracted using QIAamp Mini Kit and bisulfite treated.The IFN-γ gene promoter methylation was analyzed with methylation specific sequencing method(6 positions analyzed:-295,-186,-54, +122, +128, +171) and result was analyzed by bioinformatics software;A transwell culturing system was also established (n=6), CD4+T cells of healthy volunteers(6× 105/well)were cocultured with SPC-A1(2 × 105/well) as experimental group and CD4+T cells (6 × 105/well) cultured without SPC-A1 as control group(6 wells were established for each group).
