Several recent advances in HPLC technology are being applied to problems in pharmaceutical development.Significant efforts have been directed at increasing the speed of analysis while maintaining or even improving resolution.Columns packed with sub-2-micron particles operated in typical or high pressure modes have provided speed and resolution improvements for a number of pharmaceutical applications.Monolithic columns operated at high flow rates have also been used to reduce analysis times.New stationary phases that offer possibilities for unique selectivity continue to be introduced.Columns designed for use with highly aqueous mobile phases or to retain polar compounds have found widespread application.Columns for operation in the hydrophilic interaction mode have also been used for polar compound retention and alternative selectivity.Other columns have been designed for improved stability with high pH mobile phases.Methods for classification and comparison of column selectivity have been developed.These can be use to predict whether columns are likely to have similar or different selectivities and can aid in the choice of columns for method development.Examples illustrating applications of the above advances for pharmaceutical analysis will be described.
