Opposing roles of autophagy and the tumor suppressor UVRAG in Drosophila intestinal stem cells

Stem cells are critical for the maintenance of adult tissues and organs, including the intestinal tract of mammals and Drosophila.We find that intestinal stem cells in Drosophila depend on high levels of baseline autophagic degradation.Stem cells lacking core autophagy genes fail to maintain proliferative capacity and are progressively lost, leading to increased leakage of the gut and premature death of affected animals.In contrast, loss of the putative autophagy regulator UVRAG, a tumor suppressor potentially involved in colorectal cancer, causes abnormal proliferation of stem cells and accumulation of undifferentiated progeny.The molecular mechanisms involved in these alterations will be discussed at the conference.Moreover, we find that hyperplasia induced by lack of UVRAG requires a functional autophagy pathway, which raises the possibility of applying autophagy inhibitors for treating UVRAG-deficient cancers.Our results establish the fundamental role of autophagy for gut functioning through long-term maintenance of intestinal stem cells, which is also important for longevity.
