Chinese medicine formula “Shenqi San” inhibit proliferation of human lung adenocarcinoma A549 cells

  Purpose: The main purpose of this study was to investigate the active components of the Chinese medicine formula Shenqi San (SS) by HPLC-DAD-ESI-QTOF-MS,demonstrate the anticancer mechanism of SS on human lung adenocarcinoma A549 cells by evaluating the cell proliferation and apoptosis induction.Methods: The chloroform extraction of SS (CE-SS) was extracted from SS,while HPLC-DAD-ESI-QTOF-MS assay was performed to identified components of CE-SS.MTT assay was used to quantify the proliferation of A549 cells with the treatment of CE-SS.Apoptosis analysis was carried out by detecting phosphatidylserine (PS) externalization using the Annexin V-FITC Apoptosis Detection Kits and the stained cells were analyzed with a flow cytometer.DAPI staining assay was carried out to observe morphological characteristics of apoptotic ce11s.Western blot detected the expression of important signaling proteins involved apoptosis caspase-3,-8,-9,p53,Bax and Bcl-2.Results: Eight compounds were identified through HPLC-DAD-ESI-QTOF-MS analysis and 3-pyridine carboxylic acid,barbatin C,scutebarbatine F and barbatine D may be the main compounds responsible for the antitumor of CE-SS.The CE-SS suppress the proliferation of lung cancer A549 cells in a time and dose-dependent manner.By Annexin V-FITC/PI double staining,we can see that treatment with CE-SS induced apoptosis in A549 cells.After 24h exposure to CE-SS,the expression of protein cleaved-caspase-9,cleaved-caspase-8 and cleaved-caspase-3 were activated,the expression of protein p53 increased while the expression of Bax/Bcl-2 ratio also increased.Conclusion: This study identified the eight compounds of CE-SS,and demonstrated the anticancer effect on human lung adenocarcinoma A549 cells via induction of apoptosis.

目的:优选凤丹皮最佳乙醇回流提取工艺.方法:采用L9(34)正交试验设计,选择乙醇浓度、料液比、提取时间、提取次数为考察因素,以凤丹皮中6个主要活性成分的含量作为指标,进行综合评价.结果:凤丹皮的最佳乙醇回流提取工艺为10倍量90%的乙醇,提取60min,提取2次.结论:采用多指标综合评分法考察凤丹皮乙醇回流提取工艺,指标全面,方法客观,对于规范凤丹皮的提取工艺具有一定的意义.
气血津液是脏腑正常生理活动的产物,受脏腑支配,同时它们又是人体生命活动的物质基础,其动态变化可反映机体脏腑功能的生理与病理变化,其虚实盈亏能够直接提示人体脏腑功能的盛衰,从而影响人体的生命活动.通过构建可量化的气血津液辨识模型,分析其在代谢、功能、分布等方面的变化,判断机体虚实变化,可为中医临床诊疗提供重要依据.
目的:探讨刺梨冻干粉对UUO模型大鼠肾间质纤维化的影响及其可能的作用机制.方法:将60只SD雄性大鼠,适应性喂养一周后,随机分为12只假手术组和48只手术组;手术组行单侧输尿管结扎术(UUD),假手术组只行输尿管分离,但不结扎.术后第2天,手术组随机分为模型组、刺梨大剂量组、剌梨中剂量组、氯沙坦钾组,每组12只.各组于分组当天,均以相对应的药物灌胃.灌胃14天后处死大鼠.取大鼠肾组织观察病理变化;
目的:对新疆地区艾滋病患者常见中医证型与T淋巴细胞计数进行研究,探讨两者之间是否有相关性.方法:通过对新疆地区1130例艾滋病患者中医证型调查,发现新疆地区艾滋病患者中医证型有13种,其中气阴两虚型、气虚湿阻型居多,虚证多于实证,在此13种证型中,CD4T淋巴细胞计数呈水平分布,无明显差别.结果:新疆地区艾滋病患者常见中医证型与CD4T淋巴细胞计数无相关性.
