Protein-DNA interactions play an important role in gene expression,DNA replication,maintenance of DNA integrity and DNA damage repair.Recently Krylov has developed a new method known as non-equilibrium capillary electrophoresis of equilibrium mixtures (NECEEM) to investigate the interaction of DNA and DNA binding proteins.Free protein and DNA are separated on an uncoated capillary and elution of material is monitored by UV absorbance and fluorescence detection.We are currently determining equilibrium and kinetic parameters for the formation of complexes between a range of proteins,including methyltransferases,and double stranded-DNA.
