A live cell profiling approach for real-time monitoring and profiling of small molecules compounds will be described.The non-invasive readout allows continuous monitoring of cells inside the wells providing information regarding the temporal interaction of compounds with cells.This approach was used to obtain mechanism of action of small molecule compounds and identify off-target activities.A revised version of this technology was used for monitoring pro-arrhythmic drugs on ES-derived cardiomyocytes seeded in 96 well microtiter plates.Short term effects of bERG channel and voltage-gated calcium channel blockers as well as long term cardiotoxic effects of chemotherapeutics were clearly detected.This novel device will be of utility for safety pharmacological studies.
