We prepared a novel,sustained-release ivermectin solid dispersion(IVM-SDs)with a hydrophobic matrix(hydrogenated castor oil,HCO).No IVM crystals were observed in the IVM-SDs under polarized light microscopy and scanning electron microscopy(SEM)indicating that IVM was completely dispersed in HCO.The state of the IVM in IVM-SDs was investigated by differential scanning calorimetry(DSC)and X-ray powder diffraction(XRPD).IVM molecules were completely dispersed in the carrier as an amorphous state in the IVM-SDs when the drug: carrier ratio was lower than 1:2.In addition,no chemical reaction between drug and carrier were seen in Fourier transform infrared spectroscopy(FTIR)and 1H nuclear magnetic resonance(NMR).The in vitro release study showed that the percent drug release and release rate of IVM from IVM-SDs was considerably lower than that of pure IVM and physical mixtures(PMs)indicating that the IVM-SDs exhibited a sustained release effect of IVM.It was also found that the drug-to-carrier ratio was the critical factor for drug release.This study demonstrates that the prepared IVM-SD was an excellent candidate for the preparation of a long-acting IVM injectable aqueous suspension.
