Tetrahydrobiopterin (BH4) is an essential cofactor in hydroxylation reaction of cyclic amino acids and is synthesized via the de novo pathway in a serial reaction from guanosine triphosphate (GTP).The degeneration and cell death of dopaminergic neurons in substantia nigra pars compacta is well characterized in Parkinsons disease and the availability of cofactor, BH4, to tyrosine hydroxylase is also limited in dopamine-depleted striatum.It is clearly necessary to determine the level of bioavailable BH4 in a precision method for applying suitable diagnostic evaluation of related various diseases and for screening of potential therapeutic products to modulate BH4 levels.
