食物对比格犬口服YL-0919药代动力学的影响

  目的 研究在比格犬体内食物对YL-0919药代动力学的影响.方法 5只比格犬分别禁食12 h和未禁食后口服YL-0919 1.5 mg/kg,清洗期1周.采用LC-MS/MS法测定血浆中YL-0919浓度,用DAS 2.0软件计算药动学参数.结果 比格犬分别禁食12h和未禁食后口服YL-0919后,主要药代动力学参数为:Cmax分别为(287.73±106.50)和(276.22±115.32)ng/mL: tmax均为(1.10±0.55)h;t1/2z分别为(3.28±0.85)和(2.64±1.04)h;MRT0-24分别为(4.16±0.59)和(4.36±1.75)h:AUC0-24分别为(1242.10±462.27)和(1344.00±472.69)ng.h/mL;AUC0-∞分别为(1251.23±464.03)和(1352.91±473.46)ng·h/mL; CLz/F分别为(1.41±0.74)和(1.36±0.89)1/h/kg; Vz/F分别为(6.43±2.79)和(4.40±1.19)1/kg.经配对t检验,未禁食后口服YL-0919除t1/2t降低外,其余药动学参数差异均无统计学意义.结论 在比格犬体内食物对YL-0919药代动力学无明显影响.
