Objective To investigate the influence of nicotine pretreatment on antinociception produced by tramadol in rats.Methods The analgesic effects of chronic nicotine treatment (1 mg/kg, s.c., 7 days), acute tramadol action alone (40 mg/kg, s.c.) or tramadol administration with chronic nicotine pre-treatment were evaluated by using hot water.The tail-flick latency time from 0 to 180 min at every 15 min intervals, the percentage of maximum possible effect (% MPE) at each time point and area under curve of responses (AUC) were calculated.Results Chronic nicotine treatment alone has no antinociceptive effect compared with the baseline value.In comparison with the baseline latency, tramadol significantly increased the latency time from 60 to 180 min afterthe injection (P<0.05),and the maximum increase (l.5-fold) was at 120 min.The latency time was markedly increased from 45 to 180 min by trarnadol with nicotine co-treatment (P <0.05) compared with the baseline latency.Chronic nicotine treatment respectively increased% MPE of tramadol treatment alone at 45 min (3.85-fold, P <0.05), 60 min (2.34-fold, P <0.01), 75 min (1.68-fold, P <0.05) and 90 min (1.64-fold, P <0.05).The AUC of tramadol was increased by 1.14-fold (P <0.01) after chronic nicotine treatment.Conclusions Chronic nicotine treatment enhances the analgesic effects of tramadol in acute model of pain in rats.This work supports that the therapeutic efficacy and/or toxicity oftramadol may be altered in humans exposed to nicotine, including current and passive smokers and those on nicotine replacement treatment.
