Three dimensionally controlled optical fiber light source directs neurite growth

  Laser light can affect the growth of neural tissue, but projecting light on a controllable 3 d vector to influence neurite growth has to date not been thoroughly examined.We applied a laser beam though a tapered optical fiber positioned by a 3 axis micro-manipulator controlling position and angle.The neuritis having an obvious current growth direction were selected to be observed.An optical fiber taper was positioned at an angle perpendicular to the growth direction and 45 degrees from vertical.808 nm light from a laser was directed through the fiber, emitting from the taper end.Successful direction was considered to have occurred when neurite growth direction was altered to reach a designated target that was at least 45 degrees away from its original growth direction.Whether PC12 cells or primary cultured cells neurite, some changed direction at least 45 degrees and some changed 90 degrees within 30 minutes.The results suggests guiding the growth of a neurite by directing about 800 nm laser light to the leading edge of a neurites growing region can be accomplished by controlling the position and direction in three dimensional space of a tapered optical fiber.
