Activation of spinal microglia and astrocytes are related to bilateral mechanical allodynia in bone

  In our previous work,a rat model of tibia bone was established by inoculating the Walker 256 rat mammary gland carcinoma cells into tibia cavity.The rat showed significant bone destruction and displayed.significant ambulatory pain and mechanical allodynia after Walker 256 inoculation.In this study,the changes of glial activity was detected.The immunohistochemical results showed that there were significant astrocyte and microglia hypertrophy in both sides of spinal dorsal horn induced by unilateral bone cancer while the oligodendrocyte showed no significant changes in this model of bone cancer.Notably,the Western Blot results showed that the level of GFAP and CD 1 lb were also significant increased in both sides of spinal dorsal horn after unilateral Walker 256 or Heat-Killed tumor cells inoculation.
