雌激素受体α(estrogen receptor,ERα)是一种类固醇激素受体,在超过60%的乳腺癌中表达.临床观察和回顾性分析均显示ERα阳性的乳腺癌更易产生化疗药物耐受.近年来ERα与乳腺癌耐药的关系已日益引起关注,但其机制复杂,亟待深入研究.DNA甲基化由DNMT1、DNMT3a和DNMT3b共同催化和维持,DNMTs的异常表达直接影响基因组DNA甲基化水平.新近的报道显示,DNA甲基化异常与乳腺癌化疗耐药显著相关;类固醇激素及其受体可能参与DNA甲基转移酶的调控和肿瘤耐药的形成.本研究探讨了ERα是否能够通过调控DNMTs改变细胞基因组的DNA甲基化水平,进而增强乳腺癌细胞对化疗药物的耐受.我们的研究结果表明,ERα/estrogene可以通过上调DNMT1和DNMT3b的表达水平改变基因组DNA甲基化状态,增强ERα(+)乳腺癌细胞对化疗药物的抗性.这一研究揭示了ERα、基因组DNA甲基化和乳腺癌耐药三者间的内在联系,可为深入认识ERα(+)乳腺癌耐药的独特性和提高乳腺癌化疗疗效提供新思路.
