  Serine proteases(SPs)are crucial proteolytic insect enzymes responsible for digestion and other processes including cell signaling and immune responses.Serine protease homologs(SPHs)lack catalytic activity but function in innate immunity.The characteristics of SPs and SPHs in the globally significant pest,diamondback moth(DBM),Plutella xylostella,have not been investigated at a genome-wide scale.In our study,a total of 83 PxSP genes and 64 PxSPH genes with 28 clusters were identified in the P.xylostella genome.Most of the genes evolved from gene duplication.Phylogenetic analysis showed that 54 SP/SPH genes belonged to lepidoptera-specific and 21 to DBM-specific genes.The expression pattern of SP and SPH genes in different developmental stages,tissues and strains suggested that most lepidoptera-specific and DBM-specific SPs and SPHs in P.xylostella were highly expressed in the midgut tissue consistent with their role in digestion.Moreover,15 SPs/SPHs were highly expressed in two resistant strains,which may be associated with insecticide tolerance.Analysis of SPs and SPHs in defence against three bacteria(Enterobacter sp,Serratia sp and Enterococcus sp)showed 5 cSP/cSPH(SP/SPH containing a clip domain)genes were strikingly up-regulated after Serratia pathogen challenge,indicating that these genes might play a key role in immunity.Phylogenetic and expression pattern analyses provide insights into PxSPs and PxSPHs,which should contribute to clarifying the biological functions of this family in the future.
