Autophagy,a process of cytosolic “renovation” through lysosomal pathway,is an evolutionally conserved machinery for cellular homeostasis and cell fate decisions.The circadian clock,also called circadian rhythms(biological rhythms with a period of approximately 24 hours),was shown to play an important role in mouse autophagy.The relations between the circadian clock and autophagy,however,are not yet completely understood.Here we show that autophagy atg4a,atg4d,lc3b,gabarapa,ulk1a and ulk1b and autophagy-related genes beclin1,bnip3 and atp6v1d are all rhythmically expressed on RNA level in zebrafish under both LD(light/dark)and DD(constant dark)conditions.,and their rhythmic expression of these genes is disrupted in the per1b mutant.Luciferase reporter assays showed that atg4a promoter activities are enhanced by co-transfection of Clock1a and Bmal1b.ChIP(Chromatin Immunoprecipitation)assays showed that Bmal1b can bind to the E’ boxes in the promoter region of atg4a.Together,these results suggest that the circadian clock plays a critical role in zebrafish autophage.
