In order to establish a proper two-dimensional gel electrophoresis(2-DE)protocol for proteomic study of buckwheat seeds,Miaoqiao No.1 seed was taken as experimental material,seed protein sample of Miaoqiao No.l prepared by three methods.Two-dimensional electrophoresis was optimized in regard to protein extraction,the suitable concentration of protein per IPG gel,repetitions of experiment and SDS-PAGE parameters.The results showed the optimized system includes:total proteins of sample extracted using Phenolic extraction method,separating the proteins with 17 cm pH 4~7 IPG strips,800 μg and focusing time of 70 000 Vh.About 1193 spots can be detected,and these proteins mainly distributed in the pH 4~7 range.With good separation of the basic protein,this will be helpful to proteomic research in buckwheat seed.
