乳铁蛋白对成骨细胞中ERK1/2、JNK1/2和p38信号通路的影响

  本研究旨在研究牛乳铁蛋白对成骨细胞MAPK信号转导通路中ERK1/2、JNK1/2和p38三条亚通路的影响.采用酶原消化法从新生SD大鼠乳鼠颅盖骨中提取培养成骨细胞,设空白对照组和低中高三个剂量浓度组,培养48h后,采用RT-PCR方法检测成骨细胞MAPK信号转导通路中ERK1/2、JNK1/2和p38三条亚通路的mRNA表达水平.结果显示:与对照组比,低浓度组(20μ g/mL)中ERK1、JNK1、JNK2和p38 mRNA的表达水平分别增加了0.51倍、0.26倍、3.17倍和0.30倍;中浓度组(100μg/mL)中,ERK1、ERK2、JNK1、JNK2和p38 mRNA的表达水平分别增加了0.58倍、0.08倍、0.78倍、2.86倍和0.30倍;而高浓度组(500μg/mL)中,ERK1、ERK2、JNK1、JNK2和p38 mRNA的表达水平分别增加了0.91倍、0.11倍、0.91倍、2.38倍和0.23倍.ERK1/2和JNK1的表达量随乳铁蛋白浓度的增加而增加,呈线性剂量关系;而JNK2和p38随着乳铁蛋白浓度的增加,表达量却降低.ERK1/2、JNK1/2和p38 mRNA的表达水平变化差异均显著.说明乳铁蛋白能够促进成骨细胞中MAPK信号转导通路中ERK1/2、JNK1/2和p38三条亚通路mRNA的表达,且与浓度有关.
