Raman-activated cell sorting (RACS) offers prospects to complement the widely applied fluorescence-activated cell sorting (FACS).However,the methods used for measuring the Raman spectrum led to acquisition time of seconds to minutes per cell,which is prohibitive for rapid cell sorting applications.The on-demand single cell trap-and-release enables RACSin a flowing modeon the microfluidics device by extending the Raman spectrum acquisition time.In contrast to earlier work using optical tweezers to trap single cell[1,2],we integrated Dielectrophoresis(DEP)electrode pair into the sorting chip to trap single cell for Raman acquisition.Single Yeast cells was trapped and released serially in 20HZ frequency by controlling the voltage applied on the electrodes pair.The sorting efficiency was validated by sorting the mixture of 50% yeast cells and polystyrene beads (5 μm) and the yeast concentration was enriched to 90%.
