大豆(Glycine max)降解组文库的MicroRNAs的启动子分析

  MicroRNAs (miRNAs)是基因组编码的小分子非编码RNAs,在生长发育、生物和非生物胁迫以及其他方面均起到重要作用.为解析MicroRNAs表达调控,本研究利用TSSP程序和PlantCARE数据库预测了来自大豆降解组文库的440 miRNA的核心启动子以及369 miRNAs的顺式作用元件.结果表明:440 miRNA中有83.86%在其上游序列中包含启动子,8.64%(38个位点)在下游序列中预测到1个启动子.核心启动子的TATA盒与转录起始位点(TSSs)的分布相似,在转录起始位点100 bp区域内分布较集中.此外,对转录起始位点5的顺式作用元件预测为miRNAs的可能功能和表达的时空性提供了线索.对miRNAs的顺式作用元件和目标基因的分析显示在miRNA的表达与生长素响应因子、赤霉素响应因子之间存在潜在的负反馈调控回路.本研究为大豆miRNAs功能和表达调控研究提供了重要信息.
