Low molecular mass (MM)chitosan with high degree of deacetylation (DDA) has excellent bioactivity including targeting drug delivery,antibacterial and encapsulation properties.Thus,to reduce MM of chitosan and increase its DDA,microwave-assisted heating extraction (UAHE) conditions on the (MM) and degree of deacetylation (DDA) were investigated in this study using three factors,three levels Box-Behnken (BB) response surface design.Selected parameters including microwave heating time,H2O2 concentration and solid-liquid ratio,which significantly affect the degree of deacetylation and molecular mass of chitosan.The results showed that the observation DDA and MM were fitted to a second-order polynomial equation using multiple regression analysis and have high coefficient of predicted value (R2 =0.965).An optimization study was performed using Derringers desired function methodology and the optimal conditions involves microwave heating time of 120 s,H2O2 concentration of 2% and solid-liquid ratio of 1∶40.This study indicated that low MM of chitosan with high DDA can be applied to drug encapsulation and releasing carrier,food antibacterial and antioxidant agents.
