AIM: Acute lung injury (ALI) is associated with excessive inflammation caused by high TNF-α concentration.We hypothesized that anti-TNF-α therapy would have beneficial effects in experimental ALI in mice via downregulating the inflammation.METHODS: LPS (5mg/Kg) was intratracheally administered to the BALB/c mice (8-10 weeks old).Before LPS treatment, mice were given TNFR-Fc intraperitoneally once at a dose of 0.4mg/Kg.Wet to dry ratio of lung and protein concentration in BALF was detected.Besides, lung histology was introduced to evaluate lung injury.The concentration of IL-1β, IL-6, IL-10, IFN-γ in BALFwere analyzed by ELISA.RESULTS: TNFR-Fc treatment decreased the score of the lung histology and protein concentration in BALF significantly (P<0.001) , but not influenced the wet to dry ratio of lung obviously.The levels of IL-6 was decreased both in BALF (P< 0.001) after TNFR-Fc treatment.The concentration of IL-10 rise in BALF slightly (P=0.147).The concentration of IL-1β and IFN-γ in BALF were not be affected obviously by TNFR-Fc treatment.CONCLUSION: TNFR-Fc significantly reduced LPS-induced ALl via IL-6 and IL-10 expression regulation.
