HPLC is a conventional separation technique widely used in the determination ofpestcides, food analysis water analysis, pharmacokinetics and protemics for its ability of the separation and quantitation of compounds in simple sample and complex mixture, by adjusting mobile phase composition, pH, the ionic strength, and additives.Whereas, with the increase of diversity and complexity of analytes, HPLC meet more challenges in the analysis of biological and environmental samples, such as in proteomic the low abundance proteins must be isolated and characterized and the low content toxic should be isolated and quantitated in environmental evaluation.Hence, extensive research efforts have been dedicated to improve detection sensitivity by developing various approaches for on-line enrichment(stacking, pre-concentration, focusing).
