  The accurate regulation of the initiation of DNA replication in eukaryotic cells is a key study in gene expression,and the research about replication initiation proteins also draws attention.The histone acetylation in H3/H4 histones is associated with DNA replication.Recent findings demonstrate that H4K5 deacetylation affects DNA replication,however,its mechanism has not been identified.In order to exploring the importance of the fifth lysines modification in H4 histone to DNA replication of S.cerevisiae,Point mutations of H3/H4 histones was used to simulate H4K5 deacetylation.The role of H4K5 acetylation in the initiation of DNA replication was detected by the expression of replication initiation proteins,which is useful for researching the role of histone modifications in gene expression.
