The adverse condition of high glucose concentration in a person with diabetes is known to impair wound healing.A novel method for microvascular endothelial cell migration and wound healing has been developed and used to determine the effect of high glucose concentration and/or low oxygen tension on endothelial cell migration.Cell migration is an essential pre-requisite to wound healing and the intracellular mechanisms that result in mediating the impaired response will be presented. The role of a specific transcription factor, hypoxia inducible factor type 1 alpha in this process and the effectiveness of anti-oxidant therapies in counteracting these effects will be discussed. The ability to deliver the anti-oxidants within a novel formulation suitable for topical application has been formulated and used within the model to demonstrate the effectiveness of this approach.
