Baicalein triggers Beclin 1-and ERK pathway-dependent autophagy in ovarian carcinoma HEY cells

  Aim Baicalein (BA), one of the major compounds isolated from the root of Scutellaria baicalensis Gerogi, is a bioactive flavonoid with various pharmacological effects.Herein, wed like to determine whether BA triggers autophagy in ovarian carcinoma HEY cells and the involved mechanisms.Methods Cell proliferation was determined by MTT assay.BA-triggered autophagy was measured by monodansylcadaverine (MDC) staining and the expression of lipid modification of light chain-3 (LC3), respectively.SiRNA and pharmacological regulators were used to study the mechanisms of BA-triggered-autophagy.Results BA inhibited the proliferation on HEY cells after 24 h treatment.Meanwhile, BA triggered the acidic vesicular organelle and enhanced the expression of LC3II in HEY cells.Inhibition of autophagy by chloroquine (CQ), an inhibitor of autophagy, the cell viability was obviously decreased whereas the expression of cleaved PARP was significantly increased, indicating that BA triggered a protective autophagy.We also found siRNA Beclin 1 obviously reversed BA-induced LC3II expression.Furthermore, pretreatment with U0126, an MEK inhibitor, or siRNA of ERK, obviously inhibited BA-induced activation of ERK as well as expression of LC3II.Conclusion BA triggers a protective autophagy via Beclin 1 and ERK dependent manners in ovarian carcinoma HEY cells.
