DNA sequencing has come a long way since the days of Sanger sequencing,and till now next generation sequencing was widely accepted as a powerful tool for scientific research.NGS allows a virtually unlimited dynamic range for gene differential expression based on read-counting methods,comparing to the microarray whose detection range is limited by background noise.The digital nature of NGS also allows an exploring power to detect the single nucleotide variants in DNA,and novel transcripts.With the advance of NGS platform and analysis tools,fmishing a microorganism genome becomes an easier job,and genome sequence is definitely essential for further function study.NGS promises to monitor the global dynamic of microorganism transcriptome and unveil the temporal and spatial variation metabolism.For example,we applied the NGS on a survey of Deinococcus radiodurans responding to oxidative stress.We found a metabolic pathway switeh under the treatment of peroxide on D.radiodurans,and the elevation of metal transporter gene expression suggested that the relationship between specific metals and oxidative stress fighting.A further mining in the NGS data,we got the 5UTR,3UTR sequences and operon structure,adding into the gene structure information.When we refer our data to the sequence and original annotation stored at NCBI,we were able to correct some errors in the sequence and refme the annotation.Furthermore,we found some novel ORFs which were not present in database.
