The aberrant proteolytic processing of amyloid precursor protein (APP) by b-secretase (BACE 1) and g-secretase is important for the generation of beta-amyloid protein in the Alzheimers disease (AD) brains.Aβ is the major component of senile plaque and may be toxic to brain cells.To test weather BACE1 expression and beta-amyloid protein production can be induced by transient brain ischemia in a continuous way,and the toxicity of Aβ to brain cells.We use Middle Cerebral Artery Occlusion (MCAO) model,Animals were euthanized at 1d,3d,7d and 14d post-ischemia.Western blot analysis investigated that BACE-1 protein began to elevate at 1day and persisted up to 14days after ischemia compared to normal (P < 0.05).The alterations of BACE 1 protein levels were accompanied by corresponding increases of Aβx-42 peptides,which demonstrated by immunohistochemistry.
