In the field of functional proteomics,activity and affinity based probes are widely used to enrich specific protein targets from complex proteomes.The strepavidin-biotin system is often used for the MS based elucidation of the protein targets of chemical probes.Disadvantages of the strepavidin-biotin system include harsh condition for elution and contamination of targets by endogenously biotinylated proteins.A chemoselective cleavable linker introduced between the biotin and the warhead can overcome these problems.Several linkers have been reported,but the synthesis is often complex.We here report on a linker that is readily synthesized and can be applied in both solution and solid phase synthesis.
