The induction of pluripotent stem cells (iPSCs) by transcriptional factors is an important way to generate cells for therapeutic applications.However, the mechanism of reprogramming is poorly understood.Reprogramming intermediates are important model to study the mechanism.Here, reprogramming intermediates in our study that derived from mouse embryonic fibroblasts by over-expressing Klf4, c-Myc, E-cadherin and Glisl, name KMEG partial reprogrammed cells (KMEG-prCs).The cells had normal karyotypes, cellular morphology and proliferation was most like mouse embryonic stem cells.They stably proliferated in ES medium consisting of MEK1/2, FGF receptor and GSK3 inhibitor, and expressed SSEA-1, Sox2, but not AP, Oct4 and Nanog.
