The p harmaceutical i ndustry is l ooking t o c ontinuous p rocessing t o e nhance p roduction e fficiency a nd product quality,in line with guidance from regulatory agencies.It is therefore necessary to define and link material quality parameters,at all stages during processing,to those of the final product.T his study shows how controlling continuous manufacturing systems generates specific granule properties which then determines key characteristics of tablets.Experimental batches of granules were manufactured using the ConsiGma-lTM continuous granulation/drying system(GEA Pharma Systems)targeting specific quality/flow characteristics but using contrasting variable process conditions(water addition,input powder feed rates and granulator screw speed).The granules were dried,milled and lubricated prior to tableting.Granulate flow properties were measured at all manufacturing stages,including gr anulated wet mass,u sing an F T4 P owder R heometer?(Freeman T echnology); t ablet s trength was evaluated using a hardness tester.Strong correlation between tablet hardness and dynamic flow properties of the granules was observed through all stages of manufacture.This study shows how it is possible to generate specific tablet properties using different combinations of process conditions,and t hat a defined granule property c an b e u sed t o p redict a ch aracteristic o f t he t ablet-a significant step towards a full Design Space specification.
