Objective To explore the effects and mechanism ofsmall interfering RNA (siRNA)specific to cox-2 gene on radiosensitivity of esophageal cancer EC9706．Methods To establish siRNA vector of cox-2 gene,to be introduced into esophageal cancer cells EC9706 by lipofectamine,to obtain stably transfected cell line by constructing G418．After the radiation of 0Gy,2Gy and 4Gy,the expression of cox-2,MMP2,Bax and Bcl-2 in cells was detected by reverse transcriptase polymerase chain reaction(RT-PCR)．The expression of Cox-2 protein,AKT protein and phosphorylation AKT(pAKT)protein is detecteld by Westem blot．Flow cytometer was employed to evaluate apoptosis of cells．
