A rapid, simple method for the determination of tyrosine, 3-nitrotyrosine (NT) and 3-chlorotyrosine (CT) in human plasma was firstly developed based on direct analysis in real-time mass spectrometry (DART-MS/MS).The method was performed in positive ionization and multiple-reaction monitoring by the transitions of m/z 182.2/136.2 (Tyr), m/z216.2/170.1 (CT) and m/z 227.2/181.1 (NT).The experimental conditions and sample preparation method were systematically optimized in order to obtain the maximum intensity.The specifity, linear ranges, LODs, relative standard deviations (RSDs) of intra-and inter-day precisions and accuracies, matrix effects were studied.In conclusion, the method is simple, rapid and reliable to analyze free NT and CT in human plasma.
