To reduce the side effects and improve the lack of clinical treatment countermeasures in endometriosis chemotherapy, we have designed the glycolipid-like micelles which formed by lipid grafted chitosan mediated the pigment epithelium derived factor (PEDF) gene delivery system for the ectopic lesions of endometriosis model animals.Due to the cationic property, the glycolipid-like micelles could compact the PEDF to form complexes nanoparticles, which can efficiently protect the condensed PEDF from enzymatic degradation. The glycolipid-like micelles with PEDF at N/P 9.6 had 135.6 nm volume average hydrodynamic diameters with a narrow size distribution and 6.4±0.1mV surface potential. PEDF can be distributed in the ectopic lesions of endometriosis model after mediated by using glycolipid-like micelles gene carrier traverse the intravenous injection. Meanwhile, the result of micelles/PEDF gene therapy is the model lesion volume inhibited significantly. In this study we have researched initial mechanism of PEDF gene therapy and explore the feasibility of the PEDF gene therapy on endometriosis. And explore new theories, new directions and new ways for the treatment of endometriosis, which enjoying important theoretical and technological innovation, vision for the clinical treatment of endometriosis disease in which provides a new approach to development.
