Cordycepin Regulates the Cytokine Expression in the Prefrontal Cortex and Hippocampus in Mice

  In recent years,a growing body of evidence has shown a close connection between the neuro-immune system,especially the cytokine expression and the pathophysiology of mood disorders.Recent studies have confirmed that cordycepin(3-deoxyadenosine,3-dA)regulates the expression levels of anti-inflammatory and pro-inflammatory cytokines in the peripheral nervous system.Here,we intraperitoneal injected the mice with two doses of cordycepin(5mg/kg,12.5mg/kg),then studied the effects of cordycepin in the central nervous system and its therapeutical target in regulating the cytokines levels in the prefrontal cortex(PFC)and hippocampus(HIP)via Western blot.We found that the expression levels of anti-inflammatory cytokines IL-4 and IL-10,as well as pro-inflammatory cytokines IL-1β and TNF-α were significantly regulated after the treatment of cordycepin.After 45-minute treatment with cordycepin,only 12.5mg/kg group showed a significant increase in IL-4 expression in the PFC.After 5-day treatment,cordycepin significantly up-regulated IL-4 and IL-10 levels in both PFC and HIP.Moreover,45-minute treatment with cordycepin did not cause significant changes in IL-1β and TNF-α expressions.However,after 5-day treatment,cordycepin significantly reduced IL-1β levels in PFC,as well as TNF-α levels in both PFC and HIP.In addition,IL-6 expression levels did not show any change after cordycepin treatment.This study provide novel evidence that cordycepin regulates cytokine expression in the central nervous system,which might offer new pharmacological mechanism(s)in the treatment of neuro-immune related diseases.
