The RIO(right open reading frame)kinases(RIOKs)include RIOK1,RIOK2 and RIOK3.Each member is implicated in 40S ribosomal subunit synthesis.Emerging evidence has suggested an important role of RIOKs in cell cycle progression,proliferation,apoptosis,migration and invasion of cancer cells.The expression profiles of RIOKs are,however,largely unknown in glioma.In the current study,quantitative real-time PCR,Western blot,and immunohistochemistry followed by cell counting were used to investigate the mRNA and protein expression of RIOKs in glioma tissues and nontumor brain tissues.We found that RIOK1 expression significantly increased at mRNA levels,but decreased at protein levels in glioma tissues compared to nontumor brain tissues.RIOK2 and RIOK3 showed consistent results at transcriptional and protein levels,both molecules significantly upregulated in glioma tissues.Overexpression of RIOK3 significantly increased the proliferation of glioma cells as determined by CCK8 and EdU assays.In addition,available database analysis revealed that low expression of RIOK1 or high expression of RIOK2 and RIOK3 significantly correlated with poor patients survival.Together with a promoting role of RIOK1 and RIOK2 in the proliferation of glioblastoma cells as found previously,these results indicate that dysregulated RIOKs levels in glioma contribute to the development and progression of this cancer,and may thus serve as therapeutic targets for glioma patients.
