Cure of cognitive disorders such as amnesia,attention deficit and Alzheimers disease is still a nightmare in the field of medicine.Our growing interest in drug-facilitated memory performance led to the development of memory enhancing agents with potential of being New Chemical Entities (NCEs).We combined prediction with synthesis and biological evaluation by taking assistance from Computer Aided Drug Discovery Program PASS which helped in designing and predicting the cognition enhancing potential of the molecules proposed to be synthesized.The series of compounds (dialkylaminoalkoxyaryl acetamides,dialkylmethylcarboxamidoaryloxy acetamides,dialkylaryloxy acetamides,dialkylnitroaryloxy acetamides,2-pyrrolidinones,diaikylaminoethoxypropanols,dialkylaminoethoxyaryl carboxamido derivatives) in PASS showed AntiAlzheimers activity with Pa>0.4 and <0.5,Pa being the probability of compounds to be active.This showed the chance of finding the activity experimentally is less but the chance to find a structurally new compound,that is,NCE was greater.However,compounds showed potent antiamnesic potential in various neuropharmacological tests (elevated plus maze test,passive avoidance test,rota rod test,writhing test,mouse abdominal constriction test) with no locomotor and muscle relaxant activity and analgesic properties.
