目的:探讨健脾益肾中药强精煎对少弱精子症模型大鼠附睾精子腺苷酸环化酶(AC)、磷酸二酯酶(PDE)基因表达影响。方法:80只成年SD大鼠按随机数字表法分成4组,正常组大鼠每天给予生理盐水,模型组给予奥硝唑(ornidazde,ORN)800mg/(kg·d),黄精赞育胶囊组给予[ORN800 mg/(kg·d)+黄精赞育胶囊400 mg/(kg·d)],强精煎组大鼠每天给予[ORN800 mg/(kg·d)+强精煎10 g/(kg·d)],4周后取各组大鼠附睾组织,实时荧光定量PCR进行AC、PDE基因表达的测定。结果:强精煎组大鼠PDE基因表达水平显著低于模型组,而AC基因表达水平显著高于模型组。结论:强精煎中药可明显下调少弱精子症模型大鼠附睾精子PDE基因表达水平并显著上调其AC基因的表达水平,可能是其治疗少弱精子症的机制之一。
Objective: To investigate the effects of Jianpi Yishen Jiangzheng Jianjing on epididymal adenylate cyclase (AC) and phosphodiesterase (PDE) gene expression in the oligoasthenozoospermic rat model. Methods: Eighty adult Sprague-Dawley rats were randomly divided into 4 groups according to random number table. The normal rats were given normal saline daily. Ornidazole (ORN) 800mg / (kg · d) (ORN 800 mg / (kg · d) + Huang Jing Zang Yu Capsule 400 mg / (kg · d)], kg · d)]. After 4 weeks, the epididymal tissues of each group were taken and the expression of AC and PDE gene was measured by real-time fluorescence quantitative PCR. Results: The strong PDR rats PDE gene expression was significantly lower than the model group, while the AC gene expression was significantly higher than the model group. CONCLUSION: Jiangjing Decoction can significantly reduce the expression of PDE gene in epididymal sperm and increase the expression of AC gene in oligoasthenozoospermic model rats, which may be one of its mechanisms in the treatment of oligoasthenospermia.
