Recently,some nanoparticles are widely used as labeling probes in biomedical assay,which mainly include quantum dots(QDs),metal clusters,gold nanoparticles(GNPs)etc.Microscopy related single particles techniques including fluorescence correlation spectroscopy(FCS),resonance light scattering correlation spectroscopy(RLSCS),single particles fluorescence trajectory analysis or blinking analysis etc.were applied for biomedical assay such as detection of biomolecules.In this paper,sensitive and homogenous sandwich detection methods were developed to measure the concentration of thrombin and DNA in biofluids based on the aptamer-induced increase of characteristic diffusion time with target biomolecules in FCS and blinking suppression of QDs.The blinking QDs were used to label the ssDNA or aptamer that specifically recognize the target DNA or thrombin.The certain experimental conditions such as the ratio of DNA to QDs etc.were systematically optimized.The methods were successfully used to directly determine the level of target biomolecules from clinical samples.The methods are free of separation,show advantages over conventional fluorescent assay methods.
