Antidepressants normalize abnormal gray matter volume in major depression disorder

  As one of the most common psychiatric disorder, major depression disorder (MDD) has been reported to have very high rates of prevalence and relapse.Although previous studies have detected brain structural abnormities in MDD, very few studies have investigated the effects of antidepressants on brain structural.This study aims to investigate whether gray matter volume (GMV) changes in MDD could be normalized in MDD subjects.Thirty seven MDD patients and thirty four healthy subjects participated in this MRI study.After 12 weeks of antidepressant treatment, eighteen of the MDD subjects who were recovered defined as having 17-item Hamilton Depression Rating Scale scores of seven or below got a second MRI scan.Voxel-based morphometry (VBM) was used to detected difference in GMV between the healthy controls, the pre-and post-treatment MDD group.Increased GMV was found in the left middle frontal gyrus (BA10), left thalamus, right middle frontal gyrus (BA9), left middle temporal gyms and left middle occipital gyrus in pre-treatment MDD patients, relative to healthy control.Meanwhile, decreased GMV in the right middle occipital gyrus, left putman, left middle frontal gyrus (BAS) and left superior frontal gyrus was obeseved in the pre-treatment MDD group.In addition, altered GMV in the left middle frontal gyrus, left middle occipital gyrus, right middle occipital gyrus and left putman were normalized in the post-treatment MDD group.But abnormal GMV in the bilateral fusiform was also detected in the recovered MDD subjects.In this study, we investigated the brain effect of antidepressants in MDD subjects using VBM.Our finding of extensively distributed GMV alterations in the MDD subjects is consistent with previous studies.We found that most of the alterations in GMV were normalized by antidepressants in recovered MDD subjects.However, we also noted that some regions still exhibited abnormal GMV in recovered MDD subjects.This study may provide insight into our understanding of MDD and the brain effects of antidepressant treatment.
