Separation of Iso-Form of Trace of Metallothioneins in Living Body Using New GFC Column and 2D-LC-IC

  There are trace level of special protein and their isoforms in living body,and these substances have important function,but detail performance is still under studying.It is effective to use 2D-LC system,combination of GFC,ion exchange and affinity columns,to separate and analyze isoform.We tried to separate isoform of trace level of metallothioneins using 2D-LC system.We used size exclusion chromatography column (0.8mmID × 250mm) as the first dimension and ion exchange mode column (0.8mmID × 50mm) as the second dimension,and ICP/MS as a detector.We have developed new high performance SEC column with micro size,because we did not satisfy the performance of conventional SEC columns.We got good results of separation of trace level of metallothionein and its isoform.
