Nuclear receptors are ligand-activated transcription factors that play important roles in normal physiology and disease development.Our work indicate that nuclear receptors are also excellent drug targets for treating GI diseases.We will present two outstanding examples here.Constitutive Androstane Receptor (CAR) is a key regulator for liver drug metabolism and bilirubin clearance.We demonstrate that CAR may be a good target for treating jaundice and drug-induced liver toxicity.Famesoid X Recetptor (FXR) is key to control cholesterol and bile acid metabolism.It is a good target for treating cholestasis,liver fibrosis and diabetes.Our recent studies also suggest that FXR is essential for liver protection and is a potential novel target for preventing hepatocellular carcinoma (HCC).
