RNA interference has showed great potential that could be utilized for treatment of many human disease.[1] The siRNA used in experiment was commonly synthesized by phosphormidte approch on a synthesizer.[2] As an alternative choice,performing the synthesis procedure in solution phase through “block” condensation strategy is suitable for scale-up synthesis of a specific sequnce if a systemic siRNA drug to be approved.[3] We synthesized siRNA targeted HBV virus gene in solution phase by phosphotriester approach and including chemical modifications.The synthesized siRNA was identified by structure and biology activity.RNAi efficiency was tested by siQuant approach with HEK293 cells.The solution-phase synthesized siRNA showed good biology activity as the solid phase synthesized counterpart.
