Objective: To investigate the neuron apoptosis and the related apoptotic factors disorder in mitochondria-mediated apoptosis pathway induced by composited Aβ in rats,and detects the effects of SSF with this model.Methods: The cerebral injuries model was established by rats received intracerebroventricular injection of composited Aβ and then accepted SSF treatment.All the rats were decapitated 60 min after the last SSF administered,and then the brain was gently separated on ice.Gima stain,immunohistochemitry,western blot,RT-PCR and immunofluorescence were used to detect the levels of Bcl-2/Bax,Bcl-xl/Bax,Cytochrome C,Apaf-1,Caspase-9/3,Endo G and AIF in mitochondria,cytochylema or nucleus.Results: The present result showed that the neuron was apoptosis and accompanied by the disturbance in which the mitochondrial membrane protein Bcl-2,Bcl-xL in cortex and hippocampus were significantly decreased and,the protein Bax,Bak,and Caspase-3 were significantly increased,the mRNA expressions of Cyt-C,Apaf-1 and Caspase-9 in cytochylema of cortex and hippocampus were marked raised,the protein level of AIF and Endo G in nucleus were also significantly increased induced by composited Aβ in rats.However,SSF can differently reverse the neuron apoptosis and the up-regulate apoptosis factors in mitochondriamediated apoptosis pathway in neuronal injured rats caused by composited Aβ.Conclusions: SSF can suppress apoptosis through inhibit the disturbance of apoptotic factor in mitochondria-mediated apoptosis pathway,which suggests that SSF,an apoptotic inhibitor maybe provide a hopeful strategy in treatment of neurodegenerative disease such as AD.
