目的:研究重组组织型纤溶酶原激活剂(rt-PA)对大鼠脑梗死溶栓治疗后脑组织中ZO-1蛋白的表达,探讨不同剂量rt-PA对脑梗死溶栓后血脑屏障的影响。方法:采用大鼠自体血栓栓塞法制备大脑中动脉闭塞动物模型,采用免疫荧光染色技术检测ZO-1蛋白的表达水平,以及比色法测定血脑屏障对伊文思蓝(evans blue,EB)的通透率。结果:与正常对照组及假手术组比较,低剂量rt-PA在溶栓治疗后脑组织中ZO-1蛋白表达升高,而且EB含量显著减低;而高剂量rt-PA组ZO-1表达降低,脑组织中EB含量显著升高。结论:低剂量rt-PA在大鼠脑梗死溶栓治疗后可能对血脑屏障存在保护作用。
Objective: To investigate the expression of ZO-1 protein in rat brain tissue after thrombolytic therapy of recombinant tissue plasminogen activator (rt-PA) in rats with cerebral infarction. To investigate the effect of rt-PA on blood-brain barrier Impact. Methods: The model of middle cerebral artery occlusion was established by autologous thromboembolism in rats. The expression of ZO-1 protein was detected by immunofluorescence staining and the effect of BBB on evans blue (EB) Permeability. Results: Compared with the normal control group and the sham operation group, the expression of ZO-1 protein in brain tissue of low-dose rt-PA group was increased and EB content was significantly decreased, while the ZO-1 expression was decreased in high-dose rt-PA group , EB content in brain tissue increased significantly. Conclusion: Low dose rt-PA may protect the blood-brain barrier after thrombolysis in rats with cerebral infarction.
