Objective: To determine the T-cell receptor repertoire Vβ gene clone type in the peripheral blood of chronic hepatitis B patients Method: Complementary DNA was synthesized from RNA which was extracted from peripheral blood mononuclear cells of chronic hepatitis B patients, amplified by RT-PCR.Clonality of each amplified product was observed by imaging through scanning by gel documentation and image analyzing system (champ gel 5550) after polyacry amide gel electrophoresis (PAGE).Result: The rate of skewed(monoclonal/oligoclonal) TCR Vβ families was higher in CD8+ T-cell subsets(35/43.9%) than in CD4+ subsets(24/29.2)(p<.01).Monoclonal TCR Vβ9 and TCR Vβ14 have been found in a relatively high frequency in CD8+ T cells and TCR Vβ1 and TCR Vβ2 in CD4+T-cells Conclusion: Our research on the analysis of TCR repertoire Vβ gene may be useful in understanding and evaluation of the immune mechanism against chronic hepatitis B virus infection.It can also be helpful to design new immunotherapic strategies for chronic hepatitis B.The finding of a relatively high frequency existence in some TCR Vβ families (TCR Vβ9, TCR Vβ14 and others) in these patients could be used in monitoring the health status of the patient
