N-methyl-D-aspartate (NMDA) receptors are ionotropic glutamate receptor subtype in the central nervous system (CNS), which is also found in peripheral non-neural tissues recently, however, the influence of activation of the NMDA receptors on the peripheral tissues is still poorly understood at present.In this study,we investigated the effect of prolonged application of NMDA on the function of pancreatic islets and β-cells in vivo and in vitro.Continuous administration of NMDA for 5 days significantly increased food intake and body weight, dramatically increased fasting plasma glucose (FPG), fasting serum insulin concentration, and significantly decreased glucose tolerance in C57/BL6 mice, while all the effects caused by NMDA were relieved by NMDA receptor antagonist MK-801.The tissue microarray analysis data suggested NMDA application significantly reduced the number of pancreatic islets per unit area, and the area of pancreatic islets in per unit pancreas area.Application of 1 mmol/L NMDA to MIN6 cells for 4 days significantly reduced the glucose stimulate insulin secretion form M1N6 β-cells.After Stimulation of 25 mM glucose, the accumulation of ATP in MIN6 cells had no change.These data suggest prolonged activation of NMDA receptors could impair the function of pancreatic islets and might play a role in promoting the injury of function of pancreatic islets.
