Electrophysiological effects of orexin on globus pallidus in rats

  Objective The globus pallidus (GP) occupies a very important position in the basal ganglia.Orexinergic system directly participates in central motor control.Morphological studies have demonstrated that orexin-1 and-2receptor were expressed in GR Methods In vivo extracellular recordings was used in the present study.Results In normal rats, micro-pressure ejection of orexin (0.01 mM) increased the spontaneous firing rate of pallidal neurons by 70.6%.Furthermore, in 6-hydroxydopamine parkinsonian rats, orexin increased the firing rate by 107.4% on the lesioned side, which was significantly stronger than that on the unlesioned side (53.6%, P<0.05), as well as that in normal rats (P<0.05).Conclusion Based on the electrophysiological actions of orexin in GP, we hypothesize that orexin neurotransmission in globus pallidus may be involved in the treatment of Parkinsons disease.
