Aim: Hepatoccllular carcinoma (HCC) has been associated to diabetes and obesity, but a possible association with the metabolic syndrome (MetS) and its potential interaction with hepatitis is open to discussion.Our previous investigations have showed that GGPPS play the critical role during hyperinsulinism.In this report, the expression and distribution of GGPPS in the liver cancer, and its clinical significance was investigated.Methods: 70 patients with hepatocellular carcinoma (HCC) were included in the study.Two different types of tissues from each HCC patient were assembled immediately after surgical resection: adjacent nonmalignant tissue within 2 cm (AT), and tissue from the tumor (TT).Normal liver tissues from 10 liver transplant donors served as healthy controls (HC) while 10 patients with liver cirrhosis as cirrhosis control (CC).The expression and distribution of GGPPS in liver and liver cancer was detected with immunohistochemistry, western blots, and real-time PCR.And the relationship between the expression of GGPPS and the clinical index was also analyzed.Results: we found that GGPPS was intensified mainly in the cytoplasm of liver tumor cells.The expression of GGPPS mRNA and protein was about five folds higher in liver cancer comparing to the adjacent non-tumor tissues.Furthermore, the expression of GGPPS is associated with cirrhosis and hepatitis, and with the biological character, such as tumor stage, differentiation, and vascular invasion, et al.And the expression of GGPPS is closely relevant to the expression of COX-2 and Ras in liver cancer.Conclusion: GGPPS is of clinical significance for liver cancer to predict the biological character of liver cancer.GGPPS might play the critical role during the development of liver cancer from fatty liver, hepatitis, or cirrhosis.
